ABSTRACT

@#[摘 要] 目的:探讨circFBXL5通过靶向miR-515-5p影响膀胱癌T24细胞的增殖、迁移、侵袭及其分子机制。方法: 收集2020年4月至2020年6月间在苏州市中西医结合医院手术切除的41例膀胱癌组织及其癌旁组织,采用qPCR法检测circFBXL5、miR-515-5p的表达;双荧光素酶报告实验验证circFBXL5与miR-515-5p之间的靶向关系,体外培养人膀胱癌T24细胞,实验分为si-NC组、si-circFBXL5组、anti-miR-NC+si-circFBXL5组和si-circFBXL5+anti-miR-515-5p组;MTT法、细胞克隆形成实验、FCM、Transwell实验和WB法分别检测转染后T24细胞的增殖、细胞克隆形成、迁移、侵袭和凋亡及BAX、Bcl-2蛋白水平。结果:膀胱癌组织中circFBXL5呈高表达,miR-515-5p呈低表达(均P<0.05);circFBXL5靶向且负向调控miR-515-5p的表达;敲减circFBXL5后T24细胞的增殖抑制率、凋亡率和BAX蛋白水平均显著增高(均P<0.05),细胞克隆形成数和迁移、侵袭细胞数均显著减少(均P<0.05),Bcl-2蛋白水平显著降低(P<0.05);同时敲减circFBXL5和miR-515-5p可部分逆转敲减circFBXL5对T24细胞增殖的抑制作用。结论:circFBXL5通过调控miR-515-5p表达影响膀胱癌T24细胞的增殖、迁移、侵袭,circFBXL5和miR-515-5p可能膀胱癌治疗的潜在分子靶标。

ABSTRACT

@#Since an epidemic occur of Corona Virus Disease 2019(COVID-19) in December, 2019, all the dental healthcare service providers in our country have been greatly impacted. The strategy of managing the dental clinics is quite different from that of the medical healthcare clinics, and the key point of the administration of those dental healthcare providers is to focus on the management of outpatient care because they only supply just a little bit of inpatient care service but quite an large amount of outpatient care service. So we think the next step is to make plausible and effective scenarios to protect our dental healthcare staff and patients against corona virus infection during the treatments procedures after the reopening all of our dental clinics. To overcome this harsh condition, the infection prevention and control strategies adopted by the Stomatological Hospital, Southern Medical University were designed to be flexible and could be adjusted promptly according to the national and local governmental orders and latest guidelines released by the Centers for Disease Control and Prevention. All these prevention procedures and protocols were customized to fit our own situation and have been updated for several times based on the latest global pandemic reports. After going through the hardest time in the past four months, it’s considered that our COVID-19 prevention rules have been proved to be efficient and work well. Further more, it has made massive progress for the hospital in improving the capability of dealing with this state of emergency, especially by previewing and triaging patients strictly to cut off the possible coronavirus spreading from the original step, enhancing the standard precautions and those specific protocols made for minimizing the droplets, aerosol and contact transmission of COVID-19 indoors. Besides, a daily supervision system was set up as a routine job and a team of qualified infection prevention specialists were assigned to check and report every incorrect details during the whole procedure of dental practice. Meanwhile, the safety and well-being of the public and our medical workers could be also guaranteed through following those detailed prevention scenarios.

ABSTRACT

@#The foreign body reaction refers to a chronic inflammatory reaction and a wound-healing reaction that mainly involve macrophages and foreign body giant cells, which occur after a biological material is implanted into the body. Since macrophages in the foreign body reaction are recruited to the surface of the material after implantation of the material, subsequent secretion of a series of inflammatory factors and fusion into foreign body giant cells may lead to the degradation of the biological materials and environmental stress cracking. Moreover, the prolongation of macrophage polarization and the influence of related receptors may also lead to the phenomenon of fiber encapsulation, resulting in poor prognosis. Some scholars are committed to reducing the response of foreign bodies from the perspective of macrophages and foreign body giant cells, specifically by regulating the secretion of related inflammatory factors, reducing the subtypes of M1 macrophages, promoting their polarization to M2 macrophages, and regulating the fusion of macrophages and selective expression of macrophage-associated receptors to regulate fibrosis. The new immunological view holds that macrophages have the potential to repair bone tissue via angioplasts and osteogenesis in foreign body reactions. Therefore, the gold standard that has long been considered in regenerative medicine, which is that an inert material does not cause a foreign body reaction, is expected to be gradually replaced by tissue engineering that regulates tissue activity and function.

ABSTRACT

Objective@#To investigate the reliability of using virtual articulator in the fabrication of zirconia crowns for tetracycline discolored teeth.@*Methods@#To treat the patient with serious tetracycline stained teeth, we use zirconia crowns to shield the stained abutment teeth. To retain the characteristics of patient′s original occlusal contacts, meanwhile, to establish a stable and evenly distributed occlusal contacts, we recorded the functional movements of the patient′s jaw before preparing the teeth and transferred this relations to a physical articulator. Afterwards, relationship of the upper and lower arches was scanned through a specified fixature. Then we can customized the fabrication of the occlusal contacts of the restorations conveniently and efficiently with virtual articulator in CAD/CAM system.@*Results@#When the final restorations were inserted , a charming smile was achieved on patient′s face with satisfactory esthetics. With the application of virtual articulator in designing process, we made evenly distributed and stable bilateral occlusal contacting spots, smooth protrusive and lateral guidance on the lingual aspects of the upper anteriors. One year follow-up, this patient was observed in good oral hygiene, and was satisfied with her esthetic and functional outcome. The soft tissue around the restorations was assessed to be healthy and no signs of inflammation.@*Conclusion@# Applying the virtual articulator kits in CAD/CAM system in designing and fabricating the function-driven restorations could be an efficient way to obtain an satisfactory long-term outcome.

ABSTRACT

ABSTRACTPurpose:To evaluate the clinical efficacy of flexible ureteroscope (F-URS) combined with holmium laser lithotripter in treating renal calculi in horseshoe kidney.Materials and Methods:From November 2010 to December 2013, the medical history and charts of sixteen patients (mean age 42.9±11.6 years, range 26-66 years), including 13 males and 3 females were analyzed retrospectively. Mean stone burden was 29±8 mm (range 17-42 mm2). Mean stone digitized surface area (DSA) was 321±94 mm2 (range 180-538 mm2). Under spinal anesthesia in a modified lithotomy position with the head down, rigid ureteroscope was placed firstly into the ureter to reach the level of the pelvis, a zebra guide wire was inserted and following the removal of the rigid ureteroscope, an ureteral access sheath was positioned along the guide wire, then passed the URF P-5 flexible ureteroscope into the renal cavities over the guidewire. After locating the stones, holmium laser lithotripsy was performed.Results:The average operative time was 92±16 minutes (range 74-127 min.). No major complications were encountered. Ten patients obtained stone-free status with one session, four obtained stone-free status after two sessions. Single session stone-free rate was 62.5%, overall stone-free rate was 87.5%. Two patients have small residual stones in the lower pole.Conclusions:F-URS combined with holmium laser lithotripter and nitinol basket, is safe and effective in dealing with moderate stone diameter (<30 mm) in HSKs with high clearance rates and low complication rates.
